Start with ignition coils, wires and plugs if they haven't been done.

Cylinder 2 misfire code

Just purchased my first Mazda RX8 2004 I have a cylinder 2 misfire code pulled all plug wires off coil packs one at a time all coils are firing can anyone give me any suggestions where to go next

How old are the coils? Just because they are firing doesn't mean they are firing strong sparks.

New coils new plugs and new wires still have a misfire code on cylinder 2

With the ignition components out of the way...

Is the car still misfiring with flashing CEL? Unplug the battery, wait for a couple of minutes, reconnect and see if the code comes back.

Also, if you still have the factory cat, that should also be looked at. Could be clogged.

any oil in the intake? its probably a good idea to clean the maf, eccentric shaft sensor and do the 20 brake stomp. as jinx said check your cat

Help

Okay unhook the battery hooked it back up cell is still flashing and code came back I've replace the wires the plugs and the coil packs I've already checked the cat it is not clogged. I am new to these cars how do I clean the mass airflow sensor shaft

Mass Airflow Sensor is cleaned using special MAF cleaner (most parts stores will have this). Other cleaners will damage it. The MAF is just after the intake airbox. Just be sure to close it all back up nicely, you don't want air leaks there.
The Eccentric Shaft Position sensor just needs a wipe, it's at the bottom of the front of the engine. I would do the 20-time brake press exercise first, because it's easiest. Key to ON but engine off, press the brake pedal repeatedly until the oil pressure gauge sweeps on its own once. This clears the computer's long-term memory and lets it relearn the sensors from the beginning.

Does it misfire at idle? or only at high RPM? Or all the time?

Oh yeah, and make sure you don't strip the MAF threads. But if you do, you can use some JB Weld to fix it.

I think you need to get 20 presses in within 8 seconds or so. Here's a video to illustrate, including the oil needle sweep.

I have read that the oil needle sweep only happens after a certain flash of the ECU for S1. If the OP never updated the flash in his ECU, it won't have the sweep.

Okay I am still at a loss I have tried everyone's recommendations and have not succeeded with anything I have now changed the plugs, change the coil packs, change the wires, tried to brake pedal reset with no luck, and also clean the mass airflow sensor still have the flash n c e l while driving. It will randomly Flash go out drive a few miles and come back on flashing. I'm about ready to pour gas on top of it and sling a match at it it's getting very frustrating still have cylinder 2 misfire pending

This leaves one question: have you done a rotary-specific compression test?

Other than ignition failure, it's hard to imagine many reasons for just one rotor to misfire. The other possibility is just that your rotor 2 seal(s) are shot.

I am located in Rutherfordton North Carolina. I have not had a compression check done but driving the car it has great horsepower it's definitely not lacking in that area

It can feel great until a Honda Civic passes you with no effort. An engine with low compression can feel fine when you rev it up enough. That's why the cranking RPM is included in the compression test.

Here are the possible causes out of the FSM.
